Dieses Modul vermittelt praktische Kenntnisse, die zur Benutzung von Säugerzellkulturen zur Produktion von pharmakologisch bedeutsamen Wirkstoffen notwendig sind. Gegenstand des Praktikums ist die Planung, Modellierung, Optimierung und experimentellen Durchführung einer FedBatch-Kultivierung rekombinanter tierischer Zellen in Rührkesselbioreaktoren zur Produktion eines Glykoproteins. In der Auswertung sollen u.a. die Unterschiede zwischen dem vom Modell vorhergesagten und dem realen Prozessverhalten erarbeitet werden.
Ausgangspunkt der Modellierung und Optimierung des FedBatch-Prozesses sind reale Fermentationsdaten der im Praktikum verwendeten Zelllinie. Dabei kommt das Simulationsprogramm 'Berkeley Madonna' zum Einsatz.In einem weiteren Experiment lernen die Teilnehmerinnen und Teilnehmer den inclined-settler als Zellrückhaltemethode für kontinuierliche Perfusionskulturen kennen und erarbeiten den Einfluss der wichtigsten Betriebsparameter.Das Praktikum findet im Laborbereich E0 statt.
