Die Studierenden sammeln praktische Erfahrungen in:

-        der Amplifikation spezifischer DNA-Abschnitte ''in vitro mittels ''Polymerase-Kettenreaktion

-        Bedeutung der Primer und der Polarität für die Wirkung der DNA-Polymerase

-        Bedeutung der hitzeresistenten DNA-Polymerase

-        diagnostische Bedeutung der Polymerase-Kettenreaktion für den Nachweis von Genmutationen und von molekularen Polymorphismen

-        Anwendung von gentechnologischen Methoden am Beispiel von Restriktionsendonukleasen

-        Nachweis und Dokumentation von DNA-Fragmenten mittels Agarose-Gelelektrophorese


Kommentar: In den Praktikumsräumen besteht Kittelpflicht.

Eine Voraussetzung zur Teilnahme ist der Besuch und bestandener Test der Vorlesung - Sicherheitsunterweisung am 02.11.

Vorausgesetzt werden zudem die Inhalte des Praktikums-Skripts, das am 12.11. im LernraumPlus erhältlich ist und zum Download bereit gestellt wird.

Im Rahmen des Praktikums können Student*Innen die individuelle Sequenzvariante (Genotyp) des Geschmacksrezeptors TAS2R38 (G-Protein gekoppelter Rezeptor) bestimmen. Mit Hilfe von Polymerasekettenreaktion (PCR) wird ein spezifischer Genabschnitt amplifiziert. Mittels Sequenzabhängiger Spaltung mit Restriktionsendonukleasen wird das Vorkommen eines molekularen Polymorphismus nachgewiesen. Die DNA-Fragmente werden mittels DNA-Gelelektrophorese aufgetrennt und anschließend dokumentiert. Im Selbsttest wird die eigenen Fähigkeit den Bitterstoff 6-n-Propylthiouracil (PROP) zu schmecken, geprüft.
