Mit diesem Praktikum sollen die Grundlagen zum Verständnis der molekularen Genetik anhand von Bakterien theoretisch und experimentell erarbeitet werden. Aufbauend auf den in den Basis- und Aufbaumodulen vermittelten Grundkenntnissen werden in diesem Modul zunächst die bakterielle Genomorganisation und deren Regulation vertiefend behandelt. Klassische Methoden, wie Kreuzung und Selektion sowie die Charakterisierung von Mutanten werden erarbeitet. Der übrige Teil des Praktikums ist der Gentechnologie vorbehalten. Die Grundlagen zu heterologer Genexpression und gezielter Mutagenese werden vermittelt und typische Experimente zur DNA-Klonierung, Expression und Mutagenese werden durchgeführt. Die entstehenden Konstrukte werden analysiert und die Ergebnisse konventionell sowie computergestützt ausgewertet. Die Ergebnisse und ausgewählte Literatur sollen im Rahmen von Kurzreferaten von den Teilnehmern präsentiert werden.
Das Modul wird mit einer Klausur abgeschlossen; außerdem ist über das Praktikum ein Protokoll zu erstellen.

The aim of this practical course is to develop a theoretical and experimental understanding of molecular genetics using bacteria. Building on the basic knowledge gained in the basic and advanced modules, this module first deals in depth with bacterial genome organization and its regulation. Classical methods such as breeding and selection as well as the characterization of mutants are developed. The remaining part of the practical course is reserved for genetic engineering. The basics of heterologous gene expression and targeted mutagenesis are taught and typical experiments on DNA cloning, expression and mutagenesis are performed. The resulting constructs are analyzed and the results are evaluated using conventional and computer-assisted methods. The results and selected literature will be presented by the participants in short presentations.
The module is completed with a written examination; in addition, a protocol of the practical course must be prepared.
