391041 Einführung in die Klonierpraxis der Prokaryoten (Pr) (SoSe 2013)

Ziel dieses Praktikums ist die Vermittlung und Vertiefung von praktischen Erfahrungen und Kompetenzen im Bereich der Klonierung von prokaryotischer DNA. In diesem Praktikum sollen, aufbauend auf den theoretischen Grundlagen aus dem Studium Molekulare Biotechnologie, erste Anwendungen und Techniken erlernt werden, die für die spätere effektive Klonierung von Vektoren für die Genexpression unerlässlich sind.
Folgende Techniken sollen in dieser Lehrveranstaltung vermittelt werden:

Umgang mit der Software Clonemanger
Entwicklung von Klonierstrategien
Fehleranalytik in Klonierstrategien
Primerdesign
PCR
PCR-Mutagenese
DNA-Aufreinigung
DNA-Ligation
DNA-Fragmentanalyse
Transformation von DNA
