Abstract:

Harnessing Multi-Omics Integration: Metaproteomics-Driven Insights into the Human Gut Microbiome

Deciphering the complex molecular interplay within cellular systems is fundamental to advancing our understanding of human health, disease, and biotechnological innovation. Omics technologies, particularly metaproteomics, have become indispensable for revealing the active functional landscape of the human gut microbiome, moving beyond the genetic potential uncovered by metagenomics to provide direct evidence of microbial protein expression and function. However, extracting meaningful insights from these high-dimensional datasets necessitates robust bioinformatics workflows, encompassing meticulous metadata management, advanced proteomic identification, and integration with metabolic modeling and machine learning-driven feature extraction.

This presentation will explore the development and application of computational tools tailored for metaproteomics-based microbiome research in the human gut. I will outline our strategies for harmonizing metadata, enhancing protein identification, linking proteomic outputs to metabolic pathways, and leveraging machine learning for biomarker discovery. Emphasis will be placed on the integration of multi-omics data through knowledge graphs and graph algorithms, which enable comprehensive analysis of host-microbe interactions and functional networks.

The latter part of the talk will demonstrate these approaches in the context of inflammatory bowel disease (IBD), showcasing how metaproteomics of fecal samples can identify disease-associated protein signatures and candidate biomarkers, while also illuminating the functional dysbiosis underlying disease pathogenesis. This integrative framework not only advances our capacity for biomarker discovery but also deepens our mechanistic understanding of gut microbiome contributions to human health and disease.
