Die Vorlesung zu diesem Modul gliedert sich in zwei Teile: den theoretischen Grundlagen der zellulären Signalverschaltung (Dr. R. Hoffrogge) sowie den biophysikalischen Techniken zur Detektion zellulärer Vorgänge und der Charakterisierung von Proteinen (Prof. Dr. Kristian Müller).
Es werden somit Kenntnisse der Regulation zellulärer Vorgänge in eukaryotischen Zellen vermittelt und deren Nutzung in der molekularen Biotechnologie aufgezeigt. Neben grundlegenden Mechanismen der Signalperzeption, -transduktion und -koordination werden die verschiedenen Klassen an Signalen (Hormone, Wachstumsfaktoren, Cytokine) und Rezeptoren (Enzym- / G-Protein- / Tyrosin-Kinase-gekoppelt etc.) behandelt. Parallel zu den Prinzipien der Signalweiterleitung (Protein-Phosphorylierungen / Kinasen; cAMP) und deren Vernetzung werden spezifische Signalwege und Regulationen in eukaryotischen Zellen genauer betrachtet.
Die biophysikalischen Techniken umfassen isolierte und mikroskopische Fluoreszenzmethoden (z. B. Fluoreszenz, Fluoreszenzpolarisation, Fluoreszenzkorrelation, Durchflusszytometrie) sowie besondere mikroskopische Methoden (z. B. Superresolution, AFM). Zur Charakterisierung isolierter Proteine werden Absorptionsmethoden (z. B. UV/VIS, IR, CD), Kalorimetrie (z. B. DSC, ITC) und Interaktionsanalysen (z. B. SPR) vorgestellt. Hierbei werden auch quantitative Aspekte der mikroskopischen Bildverarbeitung und Proteindenaturierung behandelt.
Im Seminar präsentieren die Teilnehmer ausgewählte Aspekte zellulärer Prozesse sowie biophysikalische Methoden und deren Anwendung aus der Literatur und diskutieren Chancen, Nutzen und Limitierungen der Techniken. "
