In diesem Modul werden molekularbiologische Techniken und konzeptionelle Strategien zur Herstellung und funktionellen Charakterisierung transgener Organismen vermittelt.

Im Praktikum werden eine komplette Klonierungsstrategie (von der Herstellung des chimären Genkonstrukts zur Transformation und der Charakterisierung der transgenen Pflanzen) durchgeführt und molekularbiologische Techniken nach aktuellen Standards angewendet.

Techniken, die bei diesem Modul vermittelt werden, sind unter anderem:

➢ Pipetten richtig reinigen und die Kunst des Pipettierens
➢ Plasmidisolation, Restriktion und Ligation von Vektoren, Herstellung chemokompetenter Bakterien
➢ Transformationen von Escherichia coli und Agrobacterium tumefaciens
➢ Methoden der Pflanzenanzucht
➢ Transformation der Modellpflanze Arabidopsis thaliana
➢ Selektion transgener Pflanzen
➢ Detektion temperaturabhängiger alternativer Spleißereignisse in Arabidopsis mittel RT-PCR
➢ Untersuchung lichtabhängiger Genexpression in der Nutzpflanze Hordeum vulgare (Gerste) mittels Immunoblotanalysen
➢ Chemilumineszenzdetektion und Quantifizierung von Proteinen

Ferner werden Hinweise und Tipps zu Präsentationstechniken gegeben. Gelegenheit zum Üben bieten das begleitende Seminar der Studierenden und das Abschlusskolloquium.
