o Sie lernen im Rahmen dieses Praktikums das Prinzip der Absorptionsphotometrie und das Lambert-Beersche Gesetz kennen.o Sie sind im Anschluss in der Lage, den Proteingehalt einer Lösung zu bestimmen und Enzymkinetiken aufzunehmen.
An diesem Praktikumstag erarbeiten wir uns die Grundlagen der Absorptionsphotometrie. Mithilfe des Lambert-Beerschen Gesetzes bestimmen Sie den Proteingehalt von Serumproben und untersuchen den Einfluss des pH-Wertes und der Enzymmenge auf die Aktivität des Enzyms ?-Galactosidase.1. Bestimmung des Proteingehaltes einer Lösung mit der Biuret-Reaktion
2. Aktivität der beta-Galactosidase (in Abhängigkeit von Inkubationszeit, Enzymmenge und pH)
