Das im Mastermodul angebotene Praktikum "Proteinreinigung" wird durch diese Vorlesung "Aufarbeitung biotechnologischer Produkte/Proteinreinigung" begleitet. Die Vorlesung ist Voraussetzung für die Praktikumsteilnahme.
Dieses Modul behandelt die Aufarbeitung biotechnologisch hergestellter Proteine aus tierischen Zellkulturen. Dabei wird die grundsätzliche Vorgehensweise zur Gewinnung des Produktes vermittelt, unter der besonderen Voraussetzung eines medizinisch/therapeutischen Einsatzes des Proteins. Begonnen wird mit grundsätzlichen Überlegungen zur Entwicklung einer Aufarbeitungsstrategie, über Methoden der Biomasseabtrennung, insbesondere im Industriemaßstab, bis hin zur Behandlung von Trennprozessen für Proteine, auf der Basis der Eigenschaften der Proteine. Neben relevanter, chromatographischer Grundtechniken werden auch Struktur und Eigenschaften unterschiedlicher stationärer Phasen und Methoden behandelt. An Beispielen ausgewählter, therapeutischer Produkte wird die Anwendung der o.a. Methoden in Aufarbeitungsprozessen vorgestellt. Ein besonderes Augenmerk wird auch auf die Maßstabsvergrößerung einzelner Prozessschritte und die Anforderungen an die Qualität der Produkte gelegt.
