Abstract:
Thyroid hormone (TH) signalling is essential for mammalian nervous system development and function. In neuronal tissues, THs exert regulatory action via binding of the biologically active hormone T3 to nuclear TH receptors (THRA, THRB) that act as ligand-induced transcription factors (TFs) controlling expression of many genes. Although the severe neurological phenotypes that result from perturbed TH function during fetal development emphasize the critical role of TH for human brain development, there is only very limited understanding how TH availability and TH signalling are regulated in fetal human brain. When hiPSC-derived cerebral organoids (hCOs) emerged as new in vitro models to study neurodevelopmental disease mechanisms, we adopted hCOs as a promising test system to model the consequences of altered TH action for human cortical neurogenesis. hCOs generated by our protocol consistently recapitulate key features of protracted human corticogenesis and permit TH perturbation studies with robust biological replication. We further leveraged the conceptual power of single cell RNA sequencing (scRNA-seq) to delineate cell type-specific TH action in differentiating cortical neurons and to model gene regulatory networks mediating T3 effects on neuronal development. These studies yielded a first single cell atlas of the T3-responsive transcriptome in developing human cortex comprising more than 1,500 differentially expressed genes (DEGs) affected by T3 treatment in a cell type-specific manner. Pathway analyses highlight a pleiotropic nature of T3 action with T3-regulated processes ranging from mitochondrial activity and cell cycling in neuronal progenitors (NPC) to migration, neurite formation, axon guidance and synaptic function of neurons. Lineage trajectory analyses further revealed an association between T3 induction of mitochondrial OXPHOS activity with shifts in the abundance of NPC subpopulations and a precocious depletion of the neural stem cell pool. Integration of organoid data with developmental trajectories of first trimester fetal human cortex identified T3 availability as a major determinant of the pace of neuronal differentiation and maturation. Collectively, our studies demonstrated the power of hCO models for modelling TH regulation of cortical neurogenesis and showed that the high resolution of single cell analyses can provide fundamental knowledge to better understand neurodevelopmental consequences of maternal hypo- and hyperthyroidism and resistance to thyroid hormone syndromes.

Forschende der Medizinischen Fakultät OWL, der drei Krankenhausträger, die gemeinsam das Universitätsklinikum OWL bilden (Evangelisches Klinikum Bethel, Klinikum Bielefeld, Klinikum Lippe) sowie forschungsinteressierte niedergelassene Ärzt*innen in OWL präsentieren Ihre aktuellen Forschungsprojekte und tauschen sich zu Ihrer Forschung und neuen Projektideen aus. Kolleg*innen anderer Fakultäten und nationaler und internationaler Institute sind als Redner*innen und/oder Diskutant*innen ebenfalls willkommen.
