Im Spezialmodul werden Beispiele biotechnologischer Verbesserungen von Pflanzen hinsichtlich der Toleranz gegenüber Überflutungsstress und anderen abiotische Stressformen aufgezeigt. Hierzu zählen die Klonierung und Modifikation von spezifischen molekularen Signalkomponenten unter Stress zu deren funktionellen Charakterisierung. Zudem führen wir Analysen hinsichtlich der phänotypischen, transkriptionellen und metabolischen Anpassungen an Stress durch. Folgende Methoden können angewendet werden: Genaktivitätsstudien mittels RNA-Extraktion, cDNA-Synthese und RT-qPCR-basierter Quantifizierung, Genaktivitätsmuster ausgewählter Stressgene mittels des GUS-Systems, Bestimmung von Proteinmengen von Signalkomponenten unter Stress, Transkriptionsfaktoraktivitäten im Protoplasten-System (Transaktivierungsassays mit Lumineszenzbasierten Reportergenen), Protein-Protein-Interaktionsstudien im heterologen System Hefe, Toleranzassays mit Arabidopsis-Mutanten, Datenanalyse und Vorbereitung eines Fachvortrags.

Jeder Teilnehmer hält einen Vortrag zu einem ausgewählten, im Praktikum behandelten Thema
