In diesem Modul werden molekularbiologische Techniken und konzeptionelle Strategien zur Herstellung und funktionellen Charakterisierung transgener Organismen vermittelt.

Im Praktikum wird eine komplette Klonierungsstrategie (von der Herstellung des chimären Genkonstrukts zur Transformation und der Charakterisierung der transgenen Pflanzen) durchgeführt.

Techniken, die bei diesem Modul vermittelt werden, sind unter anderem:
➢ In silico design einer Klonierstrategie mit Clonemanager® Software
➢ Plasmidisolation, Restriktion und Ligation von Vektoren, Herstellung chemo-kompetenter Bakterien
➢ Transformationen von Escherichia coli und Agrobacterium tumefaciens
➢ Methoden der Pflanzenanzucht
➢ Transformation der Modellpflanze Arabidopsis thaliana
➢ Selektion transgener Pflanzen
➢ Reportergen Assays
➢ Mikroskopie
➢ Kreuzen von Arabidopsis
➢ Untersuchung lichtabhängiger Genexpression in der Nutzpflanze Hordeum vulgare (Gerste) mittels Immunoblotanalysen
➢ Chemilumineszenzdetektion und Quantifizierung von Proteinen mittels der Aida® Software

Ferner werden Hinweise und Tipps zu Präsentationstechniken gegeben. Gelegenheit zum Üben bietet das Abschlusskolloquium.
