Lehrinhalte:
Im Projektmodul „Bioimaging“ werden unterschiedlichste Methoden der Mikroskopie erlernt. Zu diesen Techniken gehören die klassische Elektronenmikroskopie mit der Transmissions-elektronenmikroskopie (TEM) wie auch der Rasterelektronenmikroskopie (REM) und der dafür erforderlichen Probenpräparation. Ebenso werden moderne Techniken des „Life Cell Imagings“ eingeführt. Der Fokus liegt hier auf zellbiologischen Fragestellungen die mit fluoreszenten Fusionsproteinen bearbeitet werden. Fluoreszenzmikroskopie, Confocale Laser Scanning Mikroskopie (CLSM), sowie neue experimentelle Verfahren in Kooperation mit Arbeitsgruppen aus der Physik stehen hier im Mittelpunkt des Praktikums. Weiterhin werden Methoden der bildgebenden Massenspektrometrie vorgestellt. In allen Bereichen der Bildgebung werden die Projekte durch bioinformatische Werkzeuge unterstützt.Kompetenzen:
Ziel des Praktikums ist das Erlernen des eigenständigen Umgangs mit komplexen Versuchsapparaturen und Methodenprotokollen. Wesentliches Ziel ist es grundlegende und fortgeschrittene Techniken der Bildgebung mit Hinblick auf ihre biologische Aussage einschätzen zu lernen.
