Competencies

Dieses Spezialmodul dient der Vertiefung der Kenntnisse in der experimentellen Biotechnologie einzelliger Mikroalgen am Beispiel von C. reinhardtii. Es werden schwerpunktmäßig molekulargenetische Experimente zur gezielten Manipulation (Bioengineering) durchgeführt, die der Optimierung u. a. von Zellwachstum und Energieerzeugung dienen sollen.

  • Überexpression von Genen
  • Sensing von Genen (RNAi)
  • Quantitative PCR
  • Zufallsmutagenese und gezielte Mutagenese
  • Screenen von Mutanten mit Fluoreszenz Video Imaging
  • Kultivierung von Zelllinien in Mini-Bioreaktor
  • Bioinformatische Analysen
  • Analysen zur H2-Produktion in Mikroalgen

Content of teaching

Die einzellige Grünalge Chlamydomonas reinhardtii gilt als der molekularbiologische Modellorganismus der Pflanzen. Über die Herstellung von Zufalls-Knockout-Mutanten oder durch gezielte Manipulation von Zielgenen des komplett sequenzierten Genoms ist dieser Organismus die Basis für eine Vielzahl von biochemischen und physiologischen Untersuchungen. Die biotechnologische Verwendung von Mikroalgen ist vielseitig und beinhaltet u. a. ihre Kultivierung in Photobioreaktoren zur Herstellung von Kosmetika und als Nahrungsergänzung. Von besonderem Interesse ist jedoch zusätzlich die Eigenschaft einiger Spezies, wie C. reinhardtii, unter anaeroben Bedingungen Wasserstoff als Energieträger der Zukunft zu produzieren.

Notwendigkeit von zwei Modulteilprüfungen:
Das Portfolio (oder das Protokoll bzw. die Präsentation) dokumentiert den Ablauf der exemplarischen Versuche und interpretiert die erzielten Ergebnisse.
In der Klausur oder der mündlichen Prüfung wird dem gegenüber die Fähigkeit zur Verallgemeinerung und Einordnung in das Zusammenhangswissen geprüft.

Die erzielten Ergebnisse werden zudem als Präsentation in medialer Form veröffentlicht oder als Protokoll verschriftlicht.

In der Regel ist das Portfolio zu erbringen.
