Competencies

Erarbeitung aktueller Methoden zur biotechnologischen Produktion rekombinanter Proteine, Handhabung bioinformatischer Programme zum Design spezifischer Oligonukleotid-Primer, verschiedene Reinigungsverfahren. Entwicklung und Durchführung einer gezielten Strategie zur heterologen Expression verschiedener Proteinklassen für eine gewünschte Anwendung im analytischen oder präparativen Maßstab. Die vermittelten Verfahren finden ein breites Anwendungsfeld zur Produktion von ökonomisch interessanten Proteinen für die Medizin und in der chemischen Industrie. In Forschungslabors dient die Reinigung rekombinanter Proteine der Charakterisierung von Protein-Aktivitäten oder der Gewinnung spezifischer Antiseren. Zukünftig wird die gezielte Expression und Reinigung rekombinanter Proteine aller "open reading frames" eines Organismus im Mikro-Maßstab für "high throughput" Proteomics-Anwendungen mittels Protein-Micro-Arrays auch in der Diagnostik eine enorme Bedeutung erlangen.

Content of teaching

Die Expression rekombinanter Proteine in heterologen Systemen findet breite Anwendung zur Aufklärung der Funktion von Proteinen, die aufgrund von Genomsequenzen vorhergesagt sind. Mikroorganismen wie Escherichia coli eignen sich sehr gut als Bioreaktoren zur Gewinnung von Proteinen und Peptiden. Dabei steht ein breites Spektrum an Expressions-Systemen zur Verfügung, die jeweils für bestimmte Anwendungen optimal zugeschnitten sind. In dem Spezialmodul finden folgende Methoden Anwendung:

  • Standard-Klonierverfahren
  • Design von spezifischen Oligonukleotid-Primern
  • Transformation von E. coli
  • DNA-Sequenzierung, computergestützte Sequenzauswertung
  • Anzucht rekombinanter Mikroorganismen
  • Proteinreinigung, chromatographische Verfahren
  • Proteolytische Abspaltung von "tags"
  • Immunologische Techniken
  • Aktivitätstests

Notwendigkeit von zwei Modulteilprüfungen:
Das Portfolio (oder das Protokoll bzw. die Präsentation) dokumentiert den Ablauf der exemplarischen Versuche und interpretiert die erzielten Ergebnisse.
In der Klausur oder der mündlichen Prüfung wird dem gegenüber die Fähigkeit zur Verallgemeinerung und Einordnung in das Zusammenhangswissen geprüft.

Das Portfolio besteht aus der eigenständigen Vorbereitung auf die Kurstage durch das zur Verfügung gestellte Script und die darin angegebene Literatur sowie der Durchführung und Dokumentation der bereitgestellten Versuche. Die erzielten Ergebnisse werden zudem als Präsentation in medialer Form veröffentlicht oder als Protokoll verschriftlicht.

In der Regel ist das Portfolio zu erbringen.
